О пространственной структуре вируса гриппа: Исследование методом тритиевой планиграфии (реферат, курсовая, диплом, контрольная)
Оболочечные вирусы представлены большим числом представителей, к ним относятся значительное количество патогенных вирусов животных и человека. Это макромолекулярные многокомпонентные высокоупорядоченные биологические системы, включающие в свой состав бислойную липидную мембрану, поверхностные и внутренние белки и нуклеиновую кислоту. Весьма многообразны и механизмы их жизненного цикла, включающие проникновение в клетку, высвобождение РНП, репродукцию и др., каждый из которых сопряжен с определенными изменениями конформации и структуры как целого вириона, так и отдельных его компонентов.
М1 белокодин из основных структурных белков вируса гриппа. Исключительная функциональная значимость его в поддержании структуры вириона, репликации вируса, вовлечении его в процессы сборки и выпочковывания зрелых вирусных частиц сделали его предметом детального изучения.
К настоящему времени накоплены достаточно обширные данные по функциональному и антигенному картированию белка, его внутриклеточной локализации. Используя криоэлектронную микроскопию высокого разрешения, была изучена тонкая структура вириона, позволившая предложить современную модель мембраны активных (инфекционных) вирусных частиц с локализацией в ней М1 белка. В 1997 г. удалось закристаллизовать фрагмент М1 белка (остатки 2158) и, исходя из данных РСА и данных криоэлектронной микроскопии, предложить гипотетическую модель укладки этого фрагмента в вирионе. Необходимость в дальнейшем изучении пространственной организации как вириона вируса гриппа, так и отдельных его компонентов, в частности М1 белка, вполне очевидна.
Представлялось целесообразным использовать метод тритиевой планиграфии, как прямой экспериментальный подход, позволяющий получать из данных тритиевого мечения объекта информацию о всей его доступной поверхности, а через нее и структурную информацию.
Можно было также думать, что тритиевая планиграфия окажется весьма результативным инструментом исследования строения различных мембран и мембран-белковых систем. Отдельные положительные примеры подобного рода работ были известны, но серьезного исследования такого рода систем с помощью метода тритиевой планиграфии еще не проводилось.
Целью настоящей работы было изучение структурной организации вируса гриппа и прежде всего пространственной укладки М1 белка в составе вируса. Необходимые этапы работы включали оптимизацию метода получения основных структурных белков, проведение контрольных экспериментов по введению метки в модельные липосомы и анализ их сохранности в условиях опытов, разработку модельной системы для изучения мембран-белковых взаимодействий, получение меченных тритием вирусных препаратов в условиях сохранения их интактности и биологических свойств, определение внутримолекулярного распределения метки по полипептидной цепи М1 белка и моделирование его пространственной структуры в составе вириона.
выводы.
1. Показана возможность приложения метода тритиевой планиграфии к исследованию строения мембран и мебран-белковых систем. Произведены предварительные оценки коэффициента ослабления потока атомов трития при прохождении ими липидного бислоя.
2. Получены экспериментальные данные о топографии поверхности М1 белка в составе вируса. Анализ внутримолекулярного распределения метки по белку в сочетании с известными из РСА данными о топологии фрагмента М1 белка (остатки 2−158) и теоретическими методами предсказания топологии Сконцевого фрагмента белка позволили предложить модель укладки М1 белка в вирусе.
3. Получены предварительные оценки степени погруженности М1 белка в толще мембраны вируса.
4. Предложена экспериментальная модель для изучения мембранбелковых взаимодействий и, в частности, для изучения механизма индуцируемого низким рН взаимодействия вирусных сливающих белков и клеточных мембран. Для визуализации структурных перестроек белков использован метод тритиевой планиграфии.
5. Оптимизированы методы выделения основных структурных белков вируса гриппа.
