Мембранотропные производные белковых ингибиторов протеиназ (реферат, курсовая, диплом, контрольная)
Создание лекарственных препаратов, обладающих повышенной адсорбцией на мембране и способных проникать внутрь клетки, является одной из основных проблем фармакологии. Отвечающие этим требованиям белковые и пептидные препараты возможно получать путем модификации белков и пептидов компонентами мембраны или их производными. Анализ литературы свидетельствует о том, что ацилирование белков производными жирных кислот значительно улучшает мембранотропные свойства препаратов, а также в ряде случаев способствует более длительному циркулированию их в кровотоке.
К числу перспективных терапевтических средств относятся ингибиторы протеиназ белкового происхождения. Основной панкреатический ингибитор трипсина (BPTI) обладает широким спектром действия и способен ингибировать такие ферменты как трипсин, химотрипсин, эластазу лейкоцитов. Он нашел широкое применение в медицине при лечении системных заболеваний в случае нарушения баланса системы протеолиза и ее ингибирования, таких как гиперфибринолиз, панкреатиты, шоки и др. На его основе выпускаются такие лекарственные формы внутривенного введения как «Контрикал» и «Гордокс». Соевый ингибитор протеиназ типа Баумана — Бирк (BBI) обладает по сравнению с основным панкреатическим ингибитором протеиназ способностью одновременно ингибировать трипсин и эластазу гранулоцитов человека. BBI активно подавляет трансформацию клеток in vitro и канцерогенез in vivo. В настоящее время пероральные препараты на его основе проходят клинические испытания в качестве антиканцерогенного средства. Однако высокий терапевтический потенциал данных белков ограничен их быстрым клиренсом.
Целью работы является создание биологически активных мембранотропных препаратов BBI и BPTI путем модификации производными жирных кислот и изучение их поведения в модельных клеточных системах.
ОБЗОР ЛИТЕРАТУРЫ.
выводы.
1. Разработан метод ацилирования соевого ингибитора протеиназ типа Баумана-Бирк (ВВ1) и основного панкреатического ингибитора протеиназ (ВРТ1) производными жирных кислот в смеси органических растворителей, позволяющий препаративно получать гомогенные препараты с регулируемым числом вводимых гидрофобных остатков в молекуле белка. Получены и охарактеризованы препараты ВВ1 и ВРТ1, содержащие от 1 до 3 ацилированных аминогрупп.
2. Изучено антипротеиназное действие ацилированных производных ВВ1 и ВРТ1. Показано, что введение гидрофобных радикалов в молекулы указанных белковых ингибиторов практически не изменяет высокого сродства к трипсину и в тоже время приводит к более эффективному ингибированию химотрипсина и эластазы лейкоцитов, имеющих гидрофобный адсорбционный участок активного центра.
3. Показана возможность образования в водной среде полимерных агрегатов амфифильным поли-]Г-винилпирролидоном (М.в. 2600), содержащим концевую стеарильную группу, размером 310−540 нм. Обнаружено, что при включении в них препаратов ВВ1, размер агрегатов уменьшается с повышением концентрации и степени гидрофобности белков, а при определенном соотношении концентраций полимера и препарата (оле)2ВВ1 образуются мицеллярные частицы (-30 нм), полностью солюбилизирующие белок, и предотвращающие его инактивацию.
4. Продемонстрировано, что ацилированные производные белковых ингибиторов протеиназ лучше захватываются монослоем эпителиальных клеток кишечника (от 4 до 200 раз) и быстрее транслоцируются через монослой по сравнению с нативным белком. Величина эффекта зависит как от количества введенных гидрофобных остатков в молекулу белка, так и от степени ненасыщенности углеводородного радикала.
5. Установлено, что в отличие от нативного ВВ1, препарат (оле), ВВ1 проявляет противовирусное действие, что обусловлено его высокой антипротеиназной активностью и повышенной мукоадгезивностью.
