Влияние блокады синтеза белка в ЦНС на формирование различных видов долговременной памяти у крыс (реферат, курсовая, диплом, контрольная)
Способность к запоминанию и воспроизведению приобретенной в течение жизни информации является одной из важнейших форм адаптивного поведения организма и лежит в основе успешного существования от беспозвоночных до человека. Исследование механизмов памяти ведется в последние десятилетия с большой интенсивностью и по праву считается одной из наиболее приоритетных областей нейронауки. Изучение этого вопроса несомненно поможет не только попять многие фундаментальные принципы работы мозга, но и откроет путь к излечению нарушений памяти, связанных с такими тяжелыми психическими заболеваниями, как болезнь Альцгеймера, болезнь Паркинсопа, болезнь Гантингтона, наркомании, алкоголизм и др.
Проблеме памяти уделяется огромное внимание уже более 100 лет. Количество накопленного экспериментального материала в этой области постоянно растет. Однако множество открытий современной биологии, в особенности, касающиеся механизмов реализации генетической информации и жизнедеятельности эукариотической клетки, значительно усложняет наши представления о работе нервной системы при обеспечении высших психических функций, таких как память, и приводит к пересмотру традиционных теоретических концепций иейробиологии, а многие вопросы до сих пор остаются открытыми (Виноградова 2000; Arshavsky, 2003).
Так, унитарная теория памяти, согласно которой гиппокампальная система считалась критически необходимой для формирования всех типов памяти, сменилась новой. По современным представлениям существует две основные системы памяти: (1) декларативная (эксплицитная) память, критически зависящая от медиальных структур височной доли, включая гиппокампальную формацию и (2) недекларативная (имплицитная) память, не зависящая от структур гиппокампальной системы.
Одним из многих типов памяти, относящихся к недекларативной системе, является двигательная (процедурная) память, в которой хранится информация о способах исполнения репертуара двигательных навыков, приобретенных в течение личного опыта (Milner et al., 1998; Ito, 2000). В настоящее время вопрос о нейроанатомических и молекулярно-клеточных основах двигательной памяти мало разработан. Однако последние несколько лет ему стало уделяться значительное внимание (Brushers-Krug et al., 1996; Muellbacher et al., 2002). Тем не менее, до сих пор исследования формирования долговременной двигательной памяти проводятся в основном на людях, а экспериментальных моделей на животных явно недостаточно.
Общепринятая теория молекулярно-клеточных механизмов консолидации (формирования) долговременной памяти (т.е. перехода информации из лабильной кратковременной памяти в стабильную долговременную) гласит, что в основе этого явления лежит феномен синаптической пластичности, т. е. морфо-функционального усиления эффективности специфических сииаптических связей в ответ на изменение активности нейронов. «Это центральный постулат нейронауки» (Martin et al., 2000). Ключевую роль в этом процессе играет специфическая активация экспрессии генов и синтез белков de novo, участвующих в модификации существующих и/или образовании новых синаптических связей (Clayton, 2000). В связи с этим ингибиторы синтеза белка (антибиотики эукариот) являются одними из наиболее широко применяемых амнестических агентов в изучении механизмов формирования различных видов долговременной памяти от беспозвоночных до млекопитающих. Тем не менее в области психофармакологических исследований памяти, связанной с применением антибиотиков, остаются серьезные противоречия, которым до сих пор не найдено адекватного объяснения. Во-первых, постулируется, что для достижения амнезии необходимо вводить ингибиторы в очень высоких («сверхфизиологических») дозах, вызывающих предельно глубокое подавление синтеза белка в мозге. Во-вторых, в некоторых работах показано, что формирование долговременной памяти у беспозвоночных и позвоночных не нарушается глубоким подавлением синтеза белка в течение как минимум многих часов (Laudein et al., 1986; Shoel, Agranoff, 1972; Staubli et al., 1985; Tully, 1996; Westenberg et al., 1998). Влияние специфических ингибиторов тотального синтеза белка эукариотических клеток на пространственную память в тесте Морриса и двигательную память исследовалось в единичных работах (Meiri & Rosenblum, 1998; Chamberlein et al., 1963).
Таким образом, несмотря на довольно обширный материал, накопленный в течение многих лет в области изучения молекулярно-клеточных механизмов памяти, дальнейшее исследование влияния подавления синтеза белка на формирование различных типов долговременной памяти, и в особенности двигательной памяти остается актуальным.
выводы.
1. В ходе систематических исследований нами установлены дозы циклогексимида, вызывающие максимально возможное подавление синтеза белка в головном и спинном мозге при двусторонних введениях в боковые желудочки.
2. Влияние ингибиторов синтеза белка на формирование долговременной памяти зависит от типа и сложности поведенческой задачи. Так, долговременная двигательная память в тесте «выученного выпрыгивания из воды» отличается чрезвычайной устойчивостью к длительной блокаде синтеза белка в ЦНС, вызванной двукратным введением циклогексимида, тогда как формирование долговременной пространственной памяти при обучении в стандартном лабиринте Морриса полностью нарушается однократным введением ингибитора.
3. Формирование двигательной долговременной памяти в тесте «выученного выпрыгивания из воды» может протекать без активации трансляционного аппарата нейронов в течение как минимум 10 ч после научения.
4. Впервые показано, что циклогексимид в дозе 400 мкг/мозг вызывает гибель клеток головного мозга через 4 ч после введения.
5. Ингибиторы синтеза белка не являются универсальными инструментами для исследования формирования всех типов долговременной памяти.
